自拟补气活血方治疗气虚血瘀模型大鼠的血液学评价

Hematological evaluation of self-designed prescription for invigorating Qi and activating blood for treating Qi deficiency and blood stasis model rats

摘要: 目的 观察不同剂量补气活血方影响下气虚血瘀模型大鼠的体重及血液学变化情况。方法 选择SPF级SD大鼠180只,随机分组,每组30只,采用饥饿、劳累、寒冷的复合刺激方法复制气虚血瘀模型,其中,空白组不造模,正常进食进水,阳性组灌胃补阳还五汤(1.45 g/ml),补气活血方组:以10 ml/kg的剂量灌胃补气活血方,高、中、低三个组的药液浓度分别为1.25 g/ml、0.63 g/ml及0.31 g/ml。分别在造模20天、40天、60天后对模型大鼠进行半自动血液流变仪测血液流变学指标;全自动血凝仪测凝血四项;ELISA法测定血浆中PIVKA-Ⅱ、γ-谷氨酸羧化酶、F1+2片段含量。结果 高中低剂量的补气活血方在60天显著缩短APTT和PT(P<0.05),延长FIB(P<0.05);显著上调高中低切变率下大鼠全血粘度(P<0.05);血浆中PIVKA-Ⅱ含量显著下降(P<0.05),谷氨酸羧化酶和F1+2片段含量显著上升(P<0.05)。结论 补气活血方具有改善大鼠血液黏度及凝血功能的作用,改善肝脏功能,对低凝状态下气虚血瘀症具有很好的疗效。

Abstract: Objective: To observe the changes of body weight and hematology in the model rats with Qi deficiency and blood stasis under the influence of different doses of Buqi Huoxue Prescription. Methods: 180 SPF SD rats were selected and randomly divided into groups with 30 rats in each group.The model of Qi deficiency and blood stasis was duplicated by the combined stimulation method of hunger, fatigue and cold. The rats in the blank group were fed with normal water, while the rats in the positive group were given Buyang Huanwu Decoction (1.45g/ml), Buqi Huoxue Decoction group: rats were given Buqi Huoxue Decoction by gavage at the dose of 10ml/kg. The concentration of Buqi Huoxue Decoction in high, medium and low groups were 1.25g/ml, 0.63g/ml and 0.31g/ml respectively.The hemorheology indexes of the model rats were measured by semi-automatic hemorheology after 20 days, 40 days and 60 days of modeling.Four items of coagulation were measured by automatic blood coagulation analyzer.ELISA method was used in the determination of plasma PIVKA Ⅱ, gamma - glutamate carboxylase and F1 + 2 fragment content. Results: The APTT and PT (P<0.05) were significantly shortened by high, medium and low doses of fuqi and Huoxue Decoction at 60 days.extended FIB(P<0.05); The whole blood viscosity of rats with high, medium and low shear rate was significantly increased (P<0.05;PIVKA - Ⅱ content decreased significantly in plasma (P<0.05), the contents of glutamate carboxylase and F1+2 fragment increased significantly (P<0.05). Conclusion: The prescription for invigorating Qi and activating blood circulation can improve the blood viscosity and coagulation function of rats and improve liver function, which has a good effect on hypocoagulation and blood stasis.
