Expression and significance of Fascin1 and E-cadherin in gastric cancer

摘要: 目的

探讨Fascin1与E-cadherin在胃癌中的表达情况及与临床病理资料的关系,并分析两者的调控关系及与胃癌细胞侵袭转移的关系。

方法

免疫组化方法检测胃癌及癌旁组织中Fascin1与E-cadherin的表达情况并分析两种蛋白在胃癌中的表达与临床病理参数的关系及二者表达的相关性,进一步检测人胃癌SGC-7901细胞中Fascin1与E-cadherin的调控作用;细胞实验研究Fascin1与胃癌细胞侵袭迁移的关系。

结果

Fascin1在胃癌组织中表达显著高于癌旁组织,E-cadherin在胃癌组织中表达显著低于癌旁组织,胃癌中Fascin1与E-cadherin表达呈显著负相关,Fascin1、E-cadherin在胃癌组织中的表达均与性别、年龄无关,均与TNM分期、淋巴结转移、浸润深度相关;细胞实验中Fascin1敲低后,E-cadherin mRNA与蛋白表达水平明显增加;SGC-7901细胞中,Fascin1干扰后,细胞的侵袭迁移能力明显降低。

结论

Fascin1在胃癌中高表达,E-cadherin在胃癌中低表达,两者均与胃癌转移有关,Fascin1对E-cadherin具有负向调控作用,Fascin1可能参与了胃癌细胞EMT过程进而调节胃癌的转移。

Abstract: Objective

To investigate the expression of Fascin1 and E-cadherin in gastric cancer and the relationship with the clinicopathological data,and to analyze the regulation relationship between Fascin1 and E-cadherin and the relationshipwith the invasion and metastasis of gastric cancer cells.

Methods

The expression of Fascin1 and E-cadherin was detected in gastric cancer and adjacent tissues by immunohistochemical method,and the relationship between the expression of two proteins in gastric cancer and the clinicopathological parameters and the correlation between the two proteins expressionin gastric cancer were analyzed,and the regulatory role ofFascin1 and E-cadherin was further detected in human gastric cancer SGC-7901 cells; The relationship between Fascin1 and invasion and migration of gastric cancer cells was researched by cell experiments.

Results

Theexpression of Fascin1 in gastric cancer tissues was significantly higher than that of paracancerous tissues.The expression of E-cadherin in gastric carcinoma was significantly lower than that of paracancer tissues.The expression of Fascin1 and E-cadherin in gastric cancer was negatively correlated.The expression of Fascin1 and E-cadherin in gastric cancer tissues was not related to sex and age. All of them were associated with TNM stage,lymph node metastasis,and invasion depth. After Fascin1 interference by siRNA,the mRNA and protein expression level of E-cadherin significantly increased in cell experiments,and the invasion and migration ability of cells significantly decreased in SGC-7901 cells.

Conclusion

Fascin1 was highly expressed in gastric cancer,and the expression of E-cadherin is lower in gastric cancer.Both of them are related to metastasis of gastric cancer.Fascin1 has negative regulation on E-cadherin.Fascin1 may be involved in the EMT process of gastric cancer cells and then regulation of the metastasis of gastric cancer.
