芍药苷对脂多糖诱导SH-SY5Y细胞损伤的影响

Effects of paeoniflorin on lipopolysaccharide-induced SH-SY5Y cell

摘要: 目的

研究芍药苷(PF)对脂多糖(LPS)诱导SH-SY5Y细胞损伤的保护及其作用机制。

方法

体外培养SH-SY5Y细胞,给予芍药苷预保护24 h,再通过LPS诱导SH-SY5Y细胞损伤,通过CCK-8法测定SH-SY5Y细胞的存活率;Western blot法检测NLRP、Bax、Bcl2蛋白的表达变化。

结果

芍药苷对LPS诱导SH-SY5Y细胞损伤具有不同程度的保护作用,细胞存活率提高;LPS组NLRP3及Bax蛋白表达明显增加,Bcl2蛋白表达减少,而不同剂量的芍药苷可以降低NLRP3及Bax蛋白的表达,增加Bcl2蛋白的表达。

结论

芍药苷对LPS诱导SH-SY5Y细胞损伤具有保护作用,保护作用可能与其降低炎症、抑制细胞凋亡有关。

Abstract: Objective

To study the protective effect of paeoniflorin (PF) on the proliferation of SH-SY5Y (LPS)-induced neuroma cells (SH-SY5Y) and its mechanism.

Methods

Lipopolysaccharide induced cell injury model was prepared. The morphological changes of SH-SY5Y cells were observed by microscope. Cell viability of SH-SY5Y cells was determined by CCK-8. The expressions of NLRP3, Bax and Bcl2 in each group of cells were analyzed by blot (Western).

Results

Paeoniflorin had different protective effect on injury of SH-SY5Y cells; the expressions of NLRP3 and Bax in the LSP group increased significantly; different doses of LPS could decrease the expressions of NLRP3 and Bax and increase the expression of Bc12.

Conclusion

Paeoniflorin has protective effects on LPS induced SH-SY5Y cell injury, which may be related to the reduction of inflammation related protein expression.
