Objective: To investigate the role of transient receptor potential vanilloid 4 (TRPV4) and P38α in neuropathic pain induced by chronic compression of dorsal root ganglion (CCD) in rats. Methods: Healthy adult male Wistar rats were randomly divided into blank group, CCD group and CCD+ruthenium red group. Paw withdrawal mechaical threshold (PWMT) was measured 1 day before surgery, 1 day, 7 days, and 21 days after surgery in the blank group and CCD surgery group. The CCD+ruthenium red group were injected with ruthenium red on the 7th day after surgery, and the PWMT was measured before and after that; RT-PCR was used to detect the expressions of TRPV4 and P38α genes in the dorsal root ganglion (DRG) in each group of the rats. Results: Compared with the blank group, the PWMT of the CCD group decreased significantly on the operation side (P<0.001), and the CCD 7-day group had the most significant decrease (P<0.001). The expressions of TRPV4 and P38α genes in the ipsilateral DRG were significantly increased (P<0.001), and the CCD 7-day group had the most significant increase (P<0.001). After 2 hours of ruthenium red injection, the PWMT of the CCD group significantly increased (P<0.001), while the P38α gene expression in the ipsilateral DRG was significantly reduced (P<0.001).Conclusion: The expressions of TRPV4 and P38α genes in the DRG of the operative rats after CCD are consistent with the changes of PWMT. The TRPV4-P38α signal pathway may be involved in the pathological process of neuropathic pain caused by CCD.
