摘要:侵袭和转移是造成肝细胞肝癌(HCC)预后不良的重要因素。近年来,HCC上皮间质转化(EMT)已经被认为是HCC侵袭、转移的关键细胞学基础。尽管引起和维持EMT的分子机制并没有被完全阐明,但是转化生长因子-β(TGF-β)、磷脂酰肌醇3激酶(phosphatidyl inositol 3-kinase,PI3K)/蛋白激酶B(AKT)、细胞核转录因子-κB (NF-κB)、Wnt/β-连接素(β-catenin) 等细胞信号通路在EMT过程中的所起到的重要作用已经被证实。上述通路通过调控EMT过程直接或间接影响HCC的预后,这也为HCC的新型靶向药物的研究提供了分子基础。

Abstract:The poorprognosisof hepatocellular carcinoma (HCC) is strongly associated with invasion and metastasis. Recently, theepithelial-mesenchymal transition(EMT) has beenconfirmed to be the cytologicalfoundationof invasion and metastasis. Yet, the molecularmechanismof inducing andmaintaining EMT has not been expounded completely. However,it has been demonstrated that transforming growth factor-β(TGF-β), phosphatidyl inositol 3-kinase/ protein kinase B( PI3K/AKT),nuclear factor-κB(NF-κB),Wnt/β-catenin signaling pathways play pivotal roles in the initiation and development of EMT.These signalingpathways can affect theprognosisof HCC by regulating EMT. From a drug development perspective, these signal pathways are potential and attractive targets for HCC treatment.
