摘要:

目的分析云南汉族大肠癌与高加索人大肠癌细胞中N?myc下游调节基因(N?myc down?stream regulated gene 1, NDRG1)的差异。方法收集云南汉族大肠癌患者的腹水,进行癌细胞培养及传代,同时选择高加索人大肠癌细胞系Caco2进行培养。通过实时荧光定量PCR,在mRNA水平观察NDRG1在云南汉族及高加索人大肠癌细胞中的表达特点。结果云南汉族与高加索人大肠癌细胞NDRG1全长序列一致,云南汉族大肠癌中NDRG1 mRNA的表达明显高于高加索人。结论NDRG1基因在云南汉族及高加索人大肠癌细胞中核酸序列相同,但mRNA表达水平存在一定差异。

Abstract:

ObjectiveTo explore the difference of NDRG1 gene between the Han nationality in Yunnan and Caucasian colorectal cancer cells.MethodsAscites from Yunnan Han nationality patients with colorectal cancer were collected for cancer cell culture and passage, and Caucasian colorectal cancer cell line Caco2 was selected for culture. The expression characteristics of NDRG1 in Yunnan Han nationality and Caucasian colorectal cancer cells were observed at mRNA level by real-time fluorescence quantitative PCR.ResultsThe full-length sequence of NDRG1 in a case of Yunnan Han nationality and Caucasian colorectal cancer cells was consistent, the expression of NDRG1 mRNA in colorectal cancer of Yunnan Han nationality was significantly higher than that of Caucasian.ConclusionIt is speculated that the nucleic acid sequence of NDRG1 gene are the same in Yunnan Han nationality and Caucasian colorectal cancer cells, but the mRNA expression level is different.
