大黄浸膏的HPLC指纹图谱研究

摘要: 目的

建立不同批次大黄浸膏高效液相色谱法(high-performance liquid chromatography,HPLC)指纹图谱,评价大黄药材的质量。

方法

采用HPLC法,Thermo C18(4.9 mm × 250 mm,5 μm)为色谱柱,甲醇(B)-0.1%磷酸水(A)做流动相,梯度洗脱,流速1 mL/min,检测波长280 nm,柱温30 ℃,进样量为10 μL。

结果

通过使用中药指纹图谱相似度评价软件分析,共标定了16个共有峰,指认了芦荟大黄素、大黄酸、大黄酚、大黄素、大黄素甲醚5个化学成分。相似度为0.691 ~ 0.984,通过相似度评价结合SPSS聚类分析和主成分分析对10批大黄浸膏指纹图谱进行质量评价,并对指认的5个指标成分进行含量测定研究。

结论

建立的大黄浸膏HPLC指纹图谱准确、方法简便、专属性强,对完善大黄浸膏的质量评价具有重要意义。

Abstract: Objective

To establish the 10 batches of fingerprint of extractumrheiby high-performance liquid chromatography.

Methods

The chromatographic column was Thermo C18(4.9 mm × 250 mm, 5 μm), the flow phase was used with a methanol and 0.1% phosphoric acid, gradient elution and the flow rate was 1 mL/min, and the detection wavelength was set at 254 nm, and the column temperature was 30 ℃, and sample quantity was 10 μL.

Results

Totally 16 chromatographic peaks were recognized as common peaks in the fingerprints through the analysis of similarity software. And the components of aloe emodin, rheinemodin, chrysophanol and physcion were identified by this method with similarities at 0.691 ~ 0.98. Similarity evaluation combined with SPSS cluster analysis(CA) and Principal Component Analysis (PCA) was used to evaluate the fingerprint of 10 batches of extractumrheiand five markers were quantified.

Conclusion

The established method for the quantitative determination of fingerprint of extractumrheiis accurate, simple and specific. At the same time, it can be effectively applied to the quality control of extractumrhei.
