Zoledronic acid in the alleviation of osteoarthritis in mice by inhibiting Wnt5a/NF-κB mediated abnormal bone resorption in Subchondral bone

摘要:

目的观察唑来膦酸(zoledronic acid, ZOL)对小鼠膝骨关节炎(osteoarthritis, OA)的影响并探讨其可能的作用机制。方法将45只雄性C57BL/6J小鼠随机分为sham + 生理盐水(normal saline, NS)组、OA + NS组和OA + ZOL组,每组15只。采用内侧半月板失稳术建立小鼠膝OA模型。术后各组小鼠腹腔注射100 μg/kg的ZOL或等量的NS,每周2次,连续4周。术后4、8周,micro-CT检测软骨下骨结构,甲苯胺蓝和HE染色评估软骨退变程度。术后第2,4,6,8周,Elisa检测血清骨转换标志物。术后第4周,TRAP染色计数软骨下骨中的破骨细胞,免疫组化和Western blot检测Wnt5a、RANKL、IκBα、NF-κB的蛋白表达水平。结果OA + NS组在术后4周的骨体积分数(bone volume fraction, BV/TV)低于sham + NS组和OA + ZOL组,术后第8周, BV/TV高于sham + NS组和OA + ZOL组,差异均具有统计学意义(P< 0.001);术后第4周和第8周的改良Mankin评分均高于sham + NS组和OA + ZOL组,差异均具有统计学意义(P< 0.001);术后4周Oc.S/BS高于sham + NS组和OA + ZOL组,差异具有统计学意义(P< 0.001),Wnt5a通路相关的蛋白水平高于sham + NS组和OA + ZOL组,差异具有统计学意义(P< 0.001)。结论ZOL可抑制Wnt5a/NF-κB介导的软骨下异常骨吸收,减轻小鼠膝OA的退变程度,延缓OA进展。

Abstract:

ObjectiveTo investigate and explore the effect and mechanism of zoledronic acid (ZOL) on osteoarthritis (OA) in mice.MethodsA knee OA mouse model was designed by surgically destabilizing the medial meniscus (DMM). Forty-five male mice were randomly assigned to sham+NS, OA + NS, and OA + ZOL groups; mice were injected intraperitoneally with 100 μg/kg ZOL or NS, twice weekly for 4 weeks. After 4 and 8 weeks of OA induction, changes in subchondral bone were analyzed using micro-computed tomography, degenerated cartilage stained with Toluidine Blue, and HE was evaluated and scored. After 2, 4, 6 and 8 weeks of OA induction, biochemical markers of bone turnover in serum were detected using Elisa kits. Four weeks after surgery, TRAP-stained osteoclasts in subchondral bone were counted, Wnt5a,RANKL,IκBαand NF-κBin subchondral bone were measured using IHC, IF and Western blot.ResultsThe BV/TV of OA + NS group was lower than that of sham + NS group and sham + ZOL group at 4 weeks after surgery, and higher than that of sham + NS group and OA + ZOL group at 8 weeks after surgery (Pall < 0.001); Modified mankin score in OA + NS group was higher than that in sham + NS group and OA + ZOL group at 4 and 8 weeks after surgery (Pall < 0.001); Oc.S/BS in OA + NS group was higher than that in sham + NS group and OA + ZOL group at 4 weeks after surgery (P< 0.001), and the protein levels of Wnt5a, RANKL, and NF-κB were higher than those in sham + NS group and OA + ZOL group (P<0.001).ConclusionZOL suppresses Wnt5a/NF-κB mediated abnormal bone resorption, alleviated the degeneration degree of knee OA in mice, and delayed OA progression.
