Niacin inpromoting small intestinal RORγt+Tregs homeostasis

摘要:

目的探索烟酸对小肠RORγt+Tregs稳态的影响。方法构建抗生素导致肠道共生菌群失调模型,使用流式细胞术检测小肠固有层(small intestinal lamina propria,SILP)RORγt+Tregs的比例;向含抗生素饮水中添加烟酸,用流式细胞术检测小肠固有层RORγt+Tregs、TH17细胞的比例以及Ki67蛋白的表达情况。结果研究结果显示,抗生素处理会导致小肠肠道固有层RORγt+Treg比例显著降低,添加烟酸能够逆转这种情况,并能够显著促进小肠固有层RORγt+Treg的增殖,而对TH17细胞没有作用。结论烟酸促进RORrt+Treg的增殖可能是肠道菌群维持小肠固有层RORrt+Treg稳态可能的机制之一。

Abstract:

ObjectiveTo explore the role of niacin in regulating the homeostasis of RORγt+Tregs in small intestine.MethodsThe mice model of antibiotic-induced microbiota imbalance was established, and the proportion of RORγt+Tregs in small intestinal lamina propria (SILP) was tested by flow cytometry. Niacin was added into drinking water containing antibiotics to detect the proportion of RORγt+Tregs and TH17 cells using flow cytometry.The expression of Ki67 in RORγt+Tregs and TH17 cells was also tested.ResultsAntibiotics treatment reduces RORγt+Tregs proportion in SILP. Niacin administration restores the proportion and proliferation of RORγt+Treg,but does not affect TH17 in SILP of antibiotic-treated mice.ConclusionThe microbiota modulates the homeostasis of RORγt+Tregs in SILP by promoting the proliferation of RORγt+Treg via niacin.
