强直性脊柱炎的脂质组学研究

摘要: 目的

通过质谱联用技术对强直性脊柱炎(ankylosing spondylitis, AS)患者的血清进行脂质组学研究,寻找潜在的AS脂质生物标志物。

方法

采用超高效液相色谱-三重四级杆质谱(UPLC /MS/MS)联用技术,对61例AS患者及28例正常人的血清样本进行脂质组学研究,比较AS患者与正常人血清中的差异脂质代谢物。

结果

AS组和健康对照组(C组)的血清脂质指纹图谱存在明显差异,其中差异较明显的脂类代谢物主要是溶血性甘油磷脂酰胆碱类(Lyso-GPC)和甘油三酯类(TG);差异明显的脂类代谢物共定性鉴别出21种,与C组相比这21种差异脂质在AS组患者血清中共同表现了下降趋势,其中以TG类最为明显。

结论

以TG(54∶7)、Lyso-GPC(18∶2)为代表的21种差异脂质代谢物可能是AS的特异性脂质代谢标志物,可以为AS的临床研究提供进一步的理论基础。

Abstract: Objective

To study the lipidomics characteristics of ankylosing spondylitis (AS) by MS for the disease lipid markers.

Methods

The serum samples of 61 AS patients and 28 normal persons were studied by using the technique of ultra-high performance liquid chromatography triple quadrupole mass spectrometry (UPLC/MS/MS),and the serum lipid metabolites of patients with AS and healthy people were compared.

Results

Serum lipid fingerprint of AS patients and the healthy people were significantly different, and the obvious differences in lipid metabolites were lysophosphatidylcholine (Lyso-GPC) and glycerin trilaurate(TG); 21 kinds of lipid metabolites with obvious difference were identified. Compared with the healthy volunteers, these 21 kinds of different lipids showed a downward trend in the serum, among which TG were the most obvious.

Conclusion

The 21 different lipid metabolites represented by TG(54∶7)、Lyso-GPC(18∶2) may be the lipid metabolism markers of AS, which can provide further theoretical basis for the clinical research of AS.
