ADAMDEC1在人结直肠癌组织中的表达及临床意义

Expression of ADAMDEC1 and its clinical significance in colorectal cancer

摘要: 目的

探讨ADAMDEC1(A disintegrin and ametalloprotease domain-like decysin 1)在人结直肠癌(colorectal cancer,CRC)组织及癌旁组织中的表达及其与临床病理特征的关联性。

方法

选取江门市中心医院100例CRC患者手术中获取的组织标本,采用免疫组织化学染色(immunohistochemistry,IHC)及Western blot分别检测CRC组织及癌旁组织中ADAMDEC1的表达情况,并分析ADAMDEC1与CRC患者临床病理特征及预后的关系。

结果

ADAMDEC1在CRC组织中表达较癌旁2 cm和癌旁>10 cm组织明显升高(P =0.021,P =0.017);相关性分析提示ADAMDEC1高表达与肿瘤大小(P =0.003)、TNM分期(P =0.001)及淋巴转移(P =0.001)呈正相关;Kaplan-Meier生存分析显示ADAMDEC1高表达的CRC患者具有更低的总体生存期;Cox回归分析提示淋巴结转移及ADAMDEC1的表达水平是影响CRC患者术后总体生存率的独立危险因素。

结论

ADAMDEC1在CRC的发生、发展中发挥重要作用,具有成为CRC早期诊断的生物标志物及治疗靶点的潜力。

Abstract: Objective

To investigate the expression of ADAMDEC1 in colorectal cancer (CRC) and its relationship with clinicopathologic features.

Methods

CRC tissues and adjacent tissues were obtained from 100 patients with CRC treated in Jiangmen Central Hospital. IHC and Western blot were conducted to detect the expression of ADAMDEC1 levels.

Results

The expression of ADAMDEC1 in CRC tissues was elevated,compared with that in adjacent normal tissues (P =0.021,P =0.017). The elevated expression of ADAMDEC1 in CRC tissues was correlated with pathological grade (P =0.003),clinical stage (P =0.001) and lymphatic metastasis (P =0.001). Kaplan-Meier survival analysis showed that CRC patients with high ADAMDEC1 expression had lower overall survival. Cox regression analysis showed that lymph node metastasis and ADAMDEC1 expression were independent risk factors for overall survival of CRC patients.

Conclusion

ADAMDEC1 plays an important role in the occurrence and development of CRC,which has the potential to become a biomarker for early diagnosis and therapeutic target of CRC.
