miR-17-92基因簇在子宫内膜病变组织中的表达及临床意义

Expression of miR-17-92 gene cluster in endometrial lesions and its clinical significance

摘要: 目的

研究miR-17-92基因簇在子宫内膜癌(endometrial cancer,EC)组织中的表达及其与临床病理特征和预后的关系。

方法

采用实时荧光定量PCR(qRT-PCR)检测EC、非典型增生组织、正常子宫内膜组织中miR-17-92表达水平,分析miR-17-92与EC患者临床病理学特征及预后的相关性。

结果

miR-17-92在EC组织中相对表达量为1.489 ± 0.038,高于非典型增生及正常子宫内膜组织中的表达水平(1.046 ± 0.029、0.621 ± 0.032),差异有统计学意义(t= 5.0166,P= 0.001;t= 2.6827,P= 0.016);miR-17-92的表达在未绝经患者、Ⅰ型EC及深肌层浸润组织中高于Ⅱ型EC及浅肌层浸润组织,差异有统计学意义(P< 0.05);预后分析显示miR-17-92高表达组的患者5年生存率为33.9%,低表达组患者的5年生存率为49.5%,Log-rank检验结果显示,miR-17-92高表达患者的生存率低于低表达患者,差异有统计学意义(χ2= 4.074,P= 0.043)。

结论

miR-17-92在EC中高表达,可能参与EC发生、发展,可作为判断EC患者预后的指标之一。

Abstract: Objective

To analyze the expression of miR-17-92 gene cluster in endometrial lesions, and to provide a theoretical basis for further exploration of the pathogenesis and prognosis of endometrial cancer.

Methods

Real-time quantitative PCR (qRT-PCR) was used to detect the expression of miR-17-92 gene cluster in endometrial carcinoma, uterine atypical hyperplasia and normal endometrial tissue, and the relationship between miR-17-92 gene cluster and clinicopathological features and prognosis in patients with endometrial cancer was analyzed.

Results

The relative expression of miR-17-92 gene cluster in endometrial carcinoma tissues was 1.489 ± 0.038, which was higher than that in endometrial atypical hyperplasia and normal endometrial tissue (1.046 ± 0.029, 0.621 ± 0.032), which was tatistically significant (t= 5.0166,P= 0.001;t= 2.6827,P= 0.016); expression of the miR-17-92 gene cluster was high in non-menopausal patients, type Ⅰ endometrial cancer, and deep myometrial invasion tissues. There was a statistically significant difference between type Ⅱ endometrial carcinoma and superficial myometrial invasion (P> 0.05), which was not associated with other clinicopathological factors. Prognostic analysis showed that the 5-year survival rate was 33.9% in the group with high miR-17-92 expression, and 49.5% in the group with low miR-17-92 expression. Log-rank test showed that the survival rate in the group with high miR-17-92 expression was lower than that in the group with low miR-17-92 expression, and the difference was statistically significant (χ2= 4.074,P= 0.043).

Conclusion

The expression level of miR-17-92 gene cluster in endometrial carcinoma tissues is significantly increased, which may be involved in the occurrence and development of endometrial cancer, and it can be used as one of the indicators to judge the prognosis of patients with endometrial cancer.
