RCOR3 gene expression and prognostic analysis in glioblastoma multiforme

摘要:

目的利用生物信息学分析方法研究RCOR3基因在多形性胶质母细胞瘤中表达和预后情况。方法采集多组基因芯片数据库数据,包括UALCAN-OMICS、GEPIA、HPA、TIMER和STRING数据库中RCOR3基因与多形性胶质母细胞瘤基因芯片数据,进行表达和预后分析。结果GEO数据库中采集GSE13276数据集,共鉴定出300个差异表达基因,其中高表达172个,低表达128个,RCOR3基因在多形性胶质母细胞瘤中呈低表达。GO富集通路为染色质组装、白细胞介素-4和白细胞介素13信号通路等;KEGG富集通路为丝裂原活化蛋白激酶等。RCOR3基因与肿瘤免疫检查点基因MLH1、PVR、LAG3呈正相关,与CD40呈负相关。RCOR3基因启动子甲基化水平明显低于正常组,差异具有统计学意义(P< 0.05),并与多形性胶质母细胞瘤分期、IDH突变、1p/19q 缺失、平均年龄和预后相关。结论RCOR3低表达与多形性胶质母细胞瘤形成具有相关性,患者总生存周期和无病生存周期明显缩短。

Abstract:

ObjectiveTo explore the expression and prognosis of RCOR3 gene in glioblastoma multiforme using bioinformatics analysis methods.MethodsMultiple sets of gene chip databases, including the correlation data between RCOR3 gene and glioblastoma multiforme in UALCAN-OMICS, GEPIA, HPA, TIMER and STRING databases were collected and biological processes, signal pathways and interaction networks were analyzed.ResultsA total of 300 differentially expressed genes were identified in the GSE13276 dataset collected in the GEO database, of which 172 were highly expressed, 128 were low expressed, and the RCOR3 gene was low expressed. Go enrichment pathways were chromatin assembly, interleukin-4 and interleukin-13 signaling pathways, etc; The enrichment pathway of KEGG was mitogen activated protein kinase. RCOR3 gene was positively correlated with tumor immune checkpoint genes MLH1, PVR, LAG3, and negatively correlated with CD40. The promoter methylation level was significantly lower than that of the normal group, and was correlated with the staging of glioblastoma multiforme, IDH mutation, 1p/19q deletion, average age and prognosis, and the difference was statistically significant (P< 0.05).ConclusionThe low expression of RCOR3 is associated with the formation of glioblastoma multiforme, and the overall life cycle and disease-free generation cycle of patients are significantly shortened.
