摘要:

目的借助网络药理学技术,通过筛选蒙药翻白草活性成分作用靶点和疾病靶点,建立“成分-靶点-疾病”网络,探讨翻白草治疗2型糖尿病的作用机制,为传统蒙药寻找新的作用靶点提供理论依据,为其实验验证及药理研究奠定基础。方法利用中药系统药理数据库分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform, TCMSP)查询翻白草活性成分,获得相对应的靶蛋白,通过GeneCards数据库筛选有关2型糖尿病的相关靶点,并利用Venny软件获得翻白草活性成分和糖尿病的靶点交集;使用STRING数据库构建化合物的蛋白互作网络(protein interaction network,PPI),利用Metascape数据库进一步进行基因本体(gene ontology,GO)生物功能以及京都基因和基因组数据库(Kyoto encyclopedia of genes and genomes, KEGG)通路富集分析。结果通过TCMSP数据库查询以及条件筛选,获得5个候选活性分子,潜在靶点共104个,相关通路250条,通过GO和KEGG富集分析得出翻白草主要通过调控癌症通路,糖尿病并发症中AGE-RAGE信号通路以及脂质与动脉粥样硬化相关通路等发挥作用。结论翻白草治疗2型糖尿病并发症主要通过调控AGE-RAGE信号通路,使其具有降低血糖的作用,为进一步实验验证奠定了良好基础。

Abstract:

ObjectiveTo explore the mechanism of Mongolian medicine Fanbaicao in treatment of type 2 diabetes by network pharmacology.MethodsAll the chemical components related to Fanbaicao were obtained from the TCMSP database by network pharmacology. GeneCard database was searched to obtain the targets of type 2 diabetes, and the intersection targets of active ingredient and disease were obtained by Venny; STRING was used to perform a PPI analysis of the common targets and then construct a PPI network forFanbaicaoin the prevention and treatment of type 2 diabetes. further analysis of target genes of Fanbaicao in GO and KEGG pathway enrichment weremade by using Metascape database.ResultsThrough TCMSP and condition screening, 5 candidate active molecules, 104 potential targets and 250 related pathways were obtained. GO and KEGG enrichment analysisconclude showed that Fanbaicao was mainly involved in the pathways in cancer, AGE-RAGE signaling pathway in diabetic complications and lipid and atherosclerosis.ConclusionThe AGE-RAGE signal pathway targeted by Potentilla discolor in the treatment of type 2 diabetes complications has a good effects of regulatory pathway, which has the effect of hypoglycemic, thus laying a good foundation for further experimental verification.
