结直肠癌是全世界发病率较高的恶性肿瘤,治疗过程中易发生复发、转移,病死率高,因此分析相关生物学指标在肿瘤进展中的作用机制,对寻找有效靶标治疗结直肠癌具有重要临床意义。环氧化酶(cyclooxygenase,COX)是将花生四烯酸催化为前列腺素的关键酶,包含COX-1、COX-2两种同工酶,其中COX-2为环氧化酶的诱导型,在恶性肿瘤中呈高表达。COX-2高表达可通过促进肿瘤细胞增殖、促进肿瘤血管形成等多种途径促进肿瘤发生。本文就COX-2的结构特征、在结直肠癌中的作用及其抑制剂在肿瘤中的应用作一全面综述。
