TGF‐β信号通路与结肠癌5‐FU耐药性的发生机制

摘要:

5‐氟尿嘧啶(5‐fluorouracil, 5‐FU)是结肠癌化疗中最有效的药物之一,但临床易产生耐药性,严重影响疾病预后。转化生长因子‐β(transforming growth factor‐β,TGF‐β)信号通路参与细胞的生长、上皮间充质转化以及耐药性等生理病理过程。但结肠癌细胞与5‐FU耐药的机制以及TGF‐β信号通路在这一机制中的作用尚不明确。本文数据来源以“colon cancer、colorectal cancer、colorectal carcinoma、TGF‐β、drug resistance of 5‐FU”等为主题词在Pubmed数据库检索,并以“结肠癌、5‐FU耐药、TGF‐β”等词在中文数据库检索,时间段为2008年1月至2021年12月。纳入标准:(1)结肠癌化疗耐药机制的研究;(2)结肠癌5‐FU耐药机制的研究。排除标准:实验老旧及结果重复的文献,对42篇文献(英文39篇,中文3篇)进行梳理分析。结果发现,该通路的异常激活、下游转导机制异常以及该信号通路的下调或抑制均可影响结肠癌细胞对5‐FU的敏感性。说明TGF‐β在结肠癌5‐FU耐药机制中具有重要的作用,调控该通路,可能是治疗结肠癌5‐FU耐药性的新思路。

Abstract: Background

5-fluorouracil (5-FU) is one of the most effective drugs in colon cancer chemotherapy, but its drug resistance is easy to develop and seriously affects the prognosis of the disease. The transforming growth factor-β (TGF-β) signaling pathway is involved in the physiological and pathological processes of cell growth, epithelial mesenchymal transformation and drug resistance. However, the mechanism of 5-FU resistance in colon cancer cells and the role of TGF-β signaling pathway in this mechanism remain unclear.

Methods

searches were made in Pubmed database with themes such as colon cancer, cancer, metastasis, TGF-β, drug resistance of 5-FU. The words "colon cancer, colorectal cancer, colorectal carcinoma, TGF-β, drug resistance of 5-FU" were searched for in Chinese database from January 2008 to December 2021. Inclusion criteria were (1) study on the mechanism of chemotherapy resistance in colon cancer and (2) study on the mechanism of 5-FU resistance in colon cancer. In the exclusion criteria, 42 literatures (39 in English and 3 in Chinese) with old experiments and duplicate results were sorted and analyzed.

Results

It was found that abnormal activation of 5-FU pathway, abnormal downstream transduction mechanism and downregulation or inhibition of 5-FU pathway could affect the sensitivity of colon cancer cells to 5-FU.

Conclusion

TGF-β plays an important role in the mechanism of 5-FU resistance in colon cancer, and the regulation of this pathway may be a new idea for the treatment of 5-FU resistance in colon cancer.
