摘要:

焦亡是细胞死亡的一种形式,对免疫至关重要,它可由典型的半胱天冬酶1(cysteinyl asparagin1,caspase1)炎性小体诱导,也可由胞质脂多糖(lipopolysacchairde, LPS)激活半胱天冬酶4、半胱天冬酶5、半胱天冬酶11(caspase4、caspase5、caspase11)诱导。激活的半胱天冬酶将焦孔素(gasdermin, GSDM)蛋白家族成员切割,切割而成的N端片段可在细胞膜上成孔从而发生细胞焦亡,在此过程中伴随着炎性因子的激活与释放。近年来GSDM家族成员被广泛研究,焦亡的研究也不断深入。大量的研究表明,细胞焦亡在胃癌的发病机制和化疗中具有关键作用。药物可以诱导胃癌细胞发生焦亡的发现为胃癌化疗提供了新的思路。焦亡诱导肿瘤细胞死亡并且伴随的炎性反应可加速其死亡,但这种炎症反应也可能是把双刃剑,过度激活的炎性反应又可促进肿瘤细胞的发生发展。因此,本文就细胞焦亡及其在胃癌发生发展中的作用机制进行综述,以期为胃癌患者开发基于焦亡的新药提供思路。

Abstract:

Pyroptosis is a form of cell death that is essential for immunity. It can be induced by the typical inflammatory vesicles of cysteinyl asparagin1 (caspase1) or by cytoplasmic lipopolysaccharide (LPS) activating cysteinyl asparagin4, cysteinyl asparagin5 and cysteinyl asparagin11 (caspase4, caspase5, and caspase11). The activated caspase cleaves members of the gasdermin (GSDM) protein family, and the cleaved N-terminal fragments can pore in the cell membrane, resulting in cell pyroptosis, accompanied by the activation and release of inflammatory factors in this process. Recently, members of the GSDM family have been extensively studied, and the study of pyroptosis has also been deepening. The discovery that drugs can induce pyroptosis in gastric cancer cells provides new perspectives for the chemotherapy of gastric cancer. Pyroptosis induces tumor cell death and the accompanying inflammatory response exacerbates its death, but this inflammatory response may be a double-edged sword, and the over-activation of the inflammatory response can promote the development of tumor cells. Therefore, this article reviews the mechanism of pyroptosis and its effect in gastric cancer development, aiming to provide ideas for the development of new pyroptosis-based drugs for gastric cancer patients.
